So we can more or less rule out the possibility of modelling a kinase trimer, after the normal mode analyses that we ran on the inactive and active models turned out to be less satisfactory than those for the dimer and monomer models. Yet the dimer still does not manage to address the inadequacies of the monomer model, and is unable to account for several activating mutations of c-Met that have been reported in the literature. Either we have made a crucial oversight in the modelling, or there has been a mistake in reporting those mutations since there had been no way of structurally examining the mutations when they had first been reported. Either way, we are in the fourth week, and starting to flesh out a report with unsubstantial (so far) results. Gah.
